I should think Bill Murray in Groundhog Day.
He's not perhaps what you'd think of as the typical movie villain, granted, but in so far as comedies go, he's a fairly real villain. I like it when he punches Ned in the face.
&, of course, he's got a happy ending (spoiler!).
How the hell is Bill Murray a villain? Explain.

He starts out a villain! Why else do you think he has to repeat the same day over & over again? He's terrorizing the entire town of Punxsutawney!
So he's not a serial killer, or a scary guy with weird teeth, or a vengeful lizard enlarged by nuclear waste--it's a comedy, after all, so he can't be too creepy.
But he's pretty fucking mean.

What about Bill Murray in "Lost In Translation"? Taking advantage of a lonely young girl-not to mention doing a substandard job as whiskey spokesman.
